Ela tinha inicialmente planeado tornar-se modelo, mas disseram-lhe que não tinha altura suficiente em 5'5" para ser modelo de passerela e devia-se concentrar em anúncios publicitários/comerciais. Depois de ter recebido aulas de comercial e de representação, ela apaixonou-se pela representação e percebeu que ela preferia isso em vez de modelo. Ashley Greene formou-se no ensino médio (ensino secundário) cedo e mudou-se para Los Angeles para seguir uma carreira na representação. Desde então, ela apareceu como convidada em vários programas de televisão populares, tais como Punk'd e Crossing Jordan.

A grande oportunidade de Greene veio em 2008 quando ela foi escolhida para interpretar Alice Cullen em Twilight, um filme baseado no romance com o mesmo nome de Stephanie Meyer. Ela voltou com o papel de Alice em 2009 para para a sequela de Twilight, The Twilight Saga: New Moon, e em 2010 para The Twilight Saga: Eclipse, e voltará com o papel de novo em 2011, para The Twilight Saga: Breaking Dawn Part 1 e Part 2.

Ashley interpretou Michelle Burkham no filme dramático Skateland, que estreou em 2010 no Sundance Film Festival.

Ela reencontrou-se com o seu colega de elenco de Twilight, Kellan Lutz, no filme Warrior's Heart. Também protagoniza o filme de suspense Summer's Blood, bem como a produção de Dark Castle Entertainment, The Apparition, que começou a ser filmada a 1 de Fevereiro de 2010 e cuja estreia está marcada para 9 de Setembro de 2011. Ashley também participa no filme de comédia, Butter, e no filme LOL: Laughing Out Loud (versão americana), cujas estreias também estão marcadas para 2011.

Aparecendo nua e num biquini de BodyPainting, Greene pousou para anúncios da SoBe que decorreram na edição 2010 da Sports Ilustrated Swimsuit Edition. É também, actualmente, a embaixadora dos produtos da Mark Cosmetics, da Avon.

A People Magazine nomeou Ashley como uma das "mulheres mais bonitas do mundo de 2010". Ela recebeu o "The Icon Style Award" nos Hollywood Life's Young Hollywood Awards no ano passado. Greene foi também apresentada a um prémio da fundação PETA por representar a Avon e a sua política dos direitos dos animais. Ashley também já conta com dois prémios individuais dos Teen Choice Awards, de 2009 com o seu papel em Twilight e em 2010 com o seu papel em New Moon.

She reprised her role as Alice in the 2009 sequel to Twilight, The Twilight Saga: New Moon, 2010's The Twilight Saga: Eclipse, and will reprise the role again in 2011, in The Twilight Saga: Breaking Dawn Part 1 and Part 2.

People Magazine named Greene as one of the “World’s Most Beautiful Women of 2010?. She received “The Style Icon Award” at Hollywood Life’s Young Hollywood Awards this year. Greene was also presented an award from the PETA foundation for representing Avon and their no testing on animals policy.
